45 Bay is located in downtown Toronto in the city's Financial District. The development site is bounded by Front Street West to the north, Lake Shore Boulevard West to the south, Bay Street to the west and Yonge Street to the east. The site is located south of Union Station and directly east of the Air Canada Centre, in the City's South Core. Metrolinx bus terminal is contained within the eastern portion of the development, with direct pedestrian connectivity to Union Station to the east.

The bus terminal has access and frontage on Lake Shore Boulevard West. As a condition of the Site Plan Agreement, the developer is required to modify the curb lane on Lake Shore Boulevard West for a partially dedicated bus lane. The developer has also agreed, as a condition of Site Plan Approval to notify City staff, in advance of the anticipated date of first occupancy of the project, to enact bylaws with respect to the proposed westbound left-turn prohibition at the driveway access on Bay Street. The intent of this prohibition is to improve operations and safety by eliminating left turns onto Bay Street and have motorists exit this location with a right-turn movement.
